# NMN Dosage, half-life & reference guide

Also known as Nicotinamide Mononucleotide · Beta-NMN · NMN supplement

A direct precursor to NAD+, a molecule essential for cellular energy, DNA repair, and the activity of longevity-linked sirtuin enzymes. NAD+ falls in some tissues with age, and NMN reliably raises blood NAD+; small trials show modest, inconsistent functional effects.

- **Studied range:** 250–1000 mg
- **How often:** Daily
- **Half-life:** Short
- **Evidence:** Moderate
- **Routes:** Oral, Oral sublingual

Studied in research — not a recommendation.

## How it works.

Direct NAD+ precursor that raises blood NAD+ levels. Supports mitochondrial function, DNA repair, sirtuin activation and cellular energy metabolism (mouse and cell data). Research by David Sinclair and others has highlighted its role in aging pathways. NAD+ precursor, not a peptide — included as it is universally tracked alongside longevity peptide protocols.

**Commonly reported:** Subtle or no noticeable effect for many users, and a mild lift in daytime energy and stamina for some. Taken orally as a daily supplement — not a peptide, but universally tracked alongside longevity peptide protocols.

## Where NMN works.

- ### Energy & longevity

**In people.** NAD+ falls in some tissues with age, and NMN reliably raises blood NAD+; small trials show modest, inconsistent functional effects.

An area lights for what a peptide is used for and what people report, with the evidence beside it. It isn’t a promise that it works there.

## Week by week.

What to expect in the first 8 weeks

1. Day 1–14 RCT

   ### NAD+ levels are climbing

NMN is oral and well-tolerated — the simplest addition to any protocol. Blood NAD+ begins rising within days. Commonly reported: little to feel in the first weeks, while cellular repair machinery gets more fuel.

   The science

NMN is converted to NAD+ via NMNAT enzymes. Yoshino et al. (2021), a 10-week randomized placebo-controlled trial in 25 prediabetic women, showed NMN (250mg/day) increased blood NAD+ metabolites and improved muscle insulin sensitivity. The NAD+ increase supports PARP (DNA repair) and sirtuin (gene regulation) pathways.

2. Day 14–56 Community

   ### A mild energy lift, for some

Commonly reported a few weeks in: a mild lift in sustained energy and exercise recovery for some, while many notice little. NAD+ now fuels mitochondrial function at a higher baseline. Many pair it with exercise; the one runner trial combined the two.

   The science

NAD+ is essential for mitochondrial electron transport chain function. Liao et al. (2021) found NMN improved aerobic markers (oxygen use and power at the ventilatory thresholds) in 48 recreational runners over 6 weeks of training; in older men, Igarashi et al. (2022) saw higher blood NAD+ with small, nominal gains in gait speed and grip. The link to felt energy runs through cellular energy but hasn't been shown.

3. Any day RCT

   ### What NMN is built on

NMN has several small human RCTs showing modest benefits: muscle insulin sensitivity (Yoshino 2021), aerobic markers in runners (Liao 2021) and higher blood NAD+. A 2026 head-to-head found NMN and NR each about double blood NAD+ in two weeks, while plain nicotinamide doesn't. And it's on firmer legal footing now: the FDA confirmed NMN as a lawful dietary supplement in late 2025.

   The science

NMN bypasses the NAMPT bottleneck in the NAD+ salvage pathway. Christen et al. (Nature Metabolism 2026) compared precursors head-to-head: NMN and NR each ~doubled blood NAD+ over 14 days; nicotinamide did not. The longevity case (sirtuin activation + DNA repair) is supported by extensive animal data (Mills et al. 2016) and emerging human data. The FDA confirmed NMN's lawful dietary-supplement status on Sept 29 and Dec 2, 2025, reversing earlier uncertainty. Not a peptide but universally co-tracked.

## A typical run.

### Run notes

- **Cycle:** Most take continuously without cycling. Some cycle 3 months on / 1 off. Oral supplement — lower risk profile. Often stacked with resveratrol, which isn't in this library. Community practice — opinions vary. Not a clinical recommendation.
- **Timing:** Morning with or without food. Sublingual absorption faster.
- **Routes:** Oral, Oral sublingual (for better absorption)
- **Stacks:** MOTS-c, SS-31

## Safety.

### Common side effects

- Generally well tolerated
- Mild nausea at high doses
- Flushing (rare)
