# N-Acetyl Semax Amidate Dosage, half-life & reference guide

Also known as NA Semax Amidate · NASA peptide · Semax Amidate

A modified version of Semax: acetylation and amidation are meant to slow its breakdown, for more potency and longer action per dose. No published human or animal study has tested this form, so everything else is borrowed from parent Semax.

- **Studied range:** 100–600 mcg
- **How often:** Daily
- **Evidence:** Emerging
- **Route:** Intranasal

Studied in research — not a recommendation.

## How it works.

Acetylated and amidated form of Semax, designed to resist breakdown by the enzymes that degrade Semax; its bioavailability, potency and brain penetration have never been measured. Assumed to share Semax's mechanism. Users describe it as stronger per microgram than standard Semax and start at the lower end of the range.

**Commonly reported:** A stronger, quicker hit of focus and drive than regular Semax at a smaller dose, and irritability or poor sleep at higher doses or late in the day.

## Where N-Acetyl Semax Amidate works.

- ### Brain, nerves & sleep

**Reported by users.** Commonly reported: a stronger, quicker hit of focus and drive than regular Semax at a smaller dose.

An area lights for what a peptide is used for and what people report, with the evidence beside it. It isn’t a promise that it works there.

## Week by week.

What to expect in the first 8 weeks

1. Day 1–3 Community

   ### Start low — this one bites

Users describe N-Acetyl Semax Amidate as more potent per mcg than regular Semax and start at the low end of the range. Commonly reported: effects come online within 30–60 min of nasal dosing, with a noticeable lift in focus, motivation and verbal fluency. Most dose in the morning, since evening doses tend to disrupt sleep.

   The science

The acetyl and amide modifications at the N- and C-termini are designed to slow breakdown: aminopeptidases account for up to two-thirds of Semax breakdown in rat serum (Potaman 1993). Blood-brain barrier penetration hasn't been measured, and acetylation does change biology (it removed Semax's protective effect in a copper-toxicity cell assay; Magrì 2016). BDNF upregulation is shown for parent Semax in rats.

2. Day 3–14 Community

   ### BDNF signal accumulating

Early days for the daily pattern. Commonly reported: focus and motivation feel steady rather than spiky, and some users notice better memory retention and learning speed. Irritability or poor sleep is the usual sign of overstimulation, and users drop the dose when it shows up.

   The science

For parent Semax, a single nasal dose raised rat hippocampal BDNF 1.4-fold within hours (Dolotov 2006); whether BDNF accumulates with repeat dosing is untested. No study has tested the modified form's effects at any dose.

3. Day 14–28 Community

   ### Cycle wrap — keep it short

Most users cap at 2–4 weeks, since continuous use of any ACTH-adjacent compound past ~4 weeks isn't well-characterized, and take 2+ weeks off before the next round.

   The science

No chronic human dosing data exists for the acetylated-amidated form specifically. ACTH(4-10) analogs in general have not been studied long-term at supraphysiological exposure. The short-cycle pattern is conservative and derived from Russian clinical usage of the parent Semax.

4. Any day Community

   ### What N-Acetyl Semax Amidate is built on

The parent Semax is approved in Russia with decades of clinical use. The modified N-acetyl-amidated form is a community-popularized variant with a chemical rationale (resisting breakdown) but no direct studies in animals or people. The only published work on N-acetyl Semax is lab chemistry.

   The science

Semax itself is an ACTH(4-10) heptapeptide analog developed in Russia. Approved there for stroke recovery and cognitive indications. The N-acetyl-amidated form is assumed to share Semax's mechanism; its pharmacokinetics and safety haven't been studied, and the one biological comparison found shows acetylation changes activity (Magrì 2016).

5. With N-Acetyl Selank and Selank Community

   ### Focus + calm duo

Modified Semax handles focus and drive; modified Selank handles calm and anxiety. Common stack in the Russian-peptide community — morning Semax + midday/evening Selank, or just layered together for a balanced cognitive profile without one overpowering the other.

   The science

In rat studies Semax raises BDNF and monoamine signaling for drive/focus, and Selank acts on GABA/serotonin for anxiolysis. Both inhibit enkephalin-degrading enzymes (Kost 2001), so they partly overlap. The stack is popular in the peptide community; no combination studies exist.
