# FOXO4-DRI Dosage, half-life & reference guide

Also known as FOXO4-p53 DRI peptide · Senostatic peptide

An experimental senolytic peptide: in mice it selectively triggers the death of "zombie cells" (senescent cells) that build up with age and release inflammatory signals, by disrupting the survival switch they rely on.

- **Studied range:** 1–2 mg
- **How often:** 3× weekly
- **Half-life:** Hours
- **Evidence:** Emerging
- **Route:** SubQ injection

Studied in research — not a recommendation.

## How it works.

D-retro-inverso peptide that disrupts the FOXO4–p53 interaction in senescent cells, triggering apoptosis of senescent cells specifically. Experimental senolytic for clearing senescent cell burden.

**Commonly reported:** Fatigue and flu-like malaise on dosing days, and for some better skin or less joint stiffness in the weeks after a run. It has only been studied in mice and cells, with no human safety data, and is used in short, infrequent cycles.

## Where FOXO4-DRI works.

- ### Energy & longevity

**In animals.** D-retro-inverso peptide that disrupts the FOXO4–p53 interaction in senescent cells, triggering apoptosis of senescent cells specifically.

An area lights for what a peptide is used for and what people report, with the evidence beside it. It isn’t a promise that it works there.

## Week by week.

What to expect in the first 8 weeks

1. Day 1–7 Animal

   ### The hit-and-run protocol

FOXO4-DRI is used as a pulse, not a chronic dose — 3x/week for about two weeks, then nothing for months. Commonly reported: mild fatigue, a flu-like feeling or brief weakness in the first few days. Why isn't known, and there are no human safety data, so strong or lasting symptoms are a reason to stop and talk to a clinician.

   The science

The peptide disrupts the FOXO4–p53 interaction that senescent cells use to evade apoptosis. Once disrupted, p53 is released and drives selective apoptosis of senescent cells (not healthy ones) in cell and mouse studies. Baar et al. 2017 showed this in fast-aging and naturally aged mice, with better fitness, fur density and kidney function.

2. Day 7–21 Animal

   ### Post-clearance recovery

The active dosing window is typically 2 weeks, then you stop and let the body recover. Some users report noticeable improvements in skin quality, joint stiffness, and general "clearance" feel 2–4 weeks after stopping — the effect shows up after, not during.

   The science

In aged mice, selective senescent cell clearance produced phenotype improvements — hair density, physical activity, kidney function — measured weeks after dosing ended. This is why FOXO4-DRI is used infrequently (1–2x/year at most) — it's a pulse-and-wait compound, not a daily protocol.

3. Day 21–60 Community

   ### Hold off before the next round

Done with the cycle? Don't rush back. Senescent cell burden doesn't rebuild in weeks — the next sensible dosing window is 6–12 months out. Aggressive repeat protocols aren't supported by any data.

   The science

Senescent cell accumulation is a slow process measured across years. Repeat intervals are community convention: the mouse studies dosed in short series and didn't test repeat cycles months apart. No human pharmacokinetic or safety data exists for repeat FOXO4-DRI cycles.

   ### What FOXO4-DRI is built on

This is an experimental senolytic with a single landmark animal paper and zero human trials. The mechanism is interesting and community reports exist, but the evidence base is young. Approach as early-stage self-experimentation, not established protocol.

   The science

Baar et al. 2017 (Cell) is the foundational paper — D-retro-inverso peptide, selective senescent cell apoptosis via FOXO4-p53 disruption, reversed aging phenotypes in mice. No human safety studies, no pharmacokinetics in humans, no established dosing protocol beyond extrapolation from the rodent paper. In mouse models of pulmonary hypertension, eliminating senescent cells, including with FOXO4-DRI, worsened pulmonary hemodynamics (Born 2023), the only published harm signal so far.

### Reconstitution example

Reconstitute a 10 mg vial of FOXO4-DRI with 2 mL of bacteriostatic water and a 2 mg dose is 40 units on a U-100 syringe, about 5 doses per vial.

### Run notes

- **Cycle:** Very short cycles only — 3x/week for 1–2 weeks, infrequently (1–2x per year). Experimental compound. In mouse research it's studied beside navitoclax (ABT-263), another senolytic, which isn't in this library. Community practice — opinions vary. Not a clinical recommendation.
- **Timing:** Typically morning injection. Strict cycle limits due to senolytic mechanism.
- **Route:** SubQ injection
